染色质重塑复合体INO80调控基因表达的机制研究

Regulation of gene expression by chromatin remodeling complex INO80

  • 摘要: 真核生物基因组DNA与组蛋白形成核小体,但真核基因组仍可结合转录、复制和修复过程的一系列酶,该过程主要受ATP依赖的核小体重塑复合物的调节.核小体重塑复合物INO80具有独特的结构特征,可促进复制叉停滞后重启,并在复制叉与转录物碰撞后帮助转录的基因清除RNA聚合酶Ⅱ.此外,INO80复合物还参与DNA损伤修复并抑制与编码序列方向相反的不适当转录.这些不同功能的基础是可促进核小体滑动并进行组蛋白变异体交换的核心ATPase亚基.本文主要概述染色质重塑复合物INO80的组成及其介导的基因表达调控机制研究的最新进展,为进一步研究INO80的生物学功能以及其在发育和疾病中的作用提供基础.

     

    Abstract: Eukaryotic genomic DNA and histones together form nucleosomes, but eukaryotic genome DNA can bind a series of enzymes during gene transcription, replication and DNA repair, all can be regulated by ATP-dependent nucleosome remodeling complex. Chromatin remodeling complex INO80 promotes delayed restart of replication forks, and helps transcribed genes to clear RNA polymerase Ⅱ after replication forks collide with the transcript. In addition, INO80 is involved in repairing DNA damage and inhibiting inappropriate transcription at the promoter in the direction opposite to the coding sequence. The basis of these different functions is the core ATPase subunit,which can promote sliding of nucleosomes and exchange of histone variants. We review here the composition of INO80 and its roles in the regulation of gene expression, to gain insights on its biological functions and roles in development, health and disease.
