MAST1 Expression and Prognostic Significance in Chinese Patients with Glioma

摘要: 为了探讨微管相关丝/苏氨酸激酶1(microtubule-associated serine/threonine kinase 1,MAST1)在人脑胶质瘤组织中的表达及预后价值.本研究利用 GEPIA 2 数据库分析MAST1在不同肿瘤组织中的表达及与胶质瘤患者预后的关系.从中国脑胶质瘤基因组图谱计划(CGGA)数据库下载mRNA seq_325和mRNA seq_693转录组数据和临床数据,分析不同临床特征的胶质瘤组织中MAST1 mRNA的表达差异.利用Kaplan Meier 生存曲线和多因素Cox回归分析评价MAST1的表达对脑胶质瘤患者预后的影响.采用基因集富集分析(GSEA)软件探讨MAST1可能参与的信号通路.结果显示MAST1 mRNA在脑胶质瘤组织中低表达;其表达与患者的年龄、WHO分级、IDH突变、1p/19q 共缺失状态有关.MAST1低表达,脑胶质瘤患者预后较差.进一步富集分析结果显示MAST1在多种生物学过程中发挥重要作用.可见,MAST1是脑胶质瘤患者预后评估的潜在指标,本研究为脑胶质瘤的发病机制奠定了理论基础.Abstract:To investigate the expression and prognostic significance of microtubule-associated serine/threonine kinase 1 (MAST1) in glioma tissues. MAST1 mRNA expression in tumor tissues and prognosis of patients with gliomas were analyzed by the GEPIA 2 database. mRNA seq_325 and mRNA seq_693 transcriptome data and clinical data were obtained from the Chinese Glioma Genome Atlas (CGGA) database, and the differential expression of MAST1 mRNA in glioma tissues with different clinical features was analyzed. The impact of MAST1 expression on the prognosis of patients with glioma was evaluated using Kaplan-Meier survival curves and multivariate Cox regression analysis. The potential signaling pathways involved in MAST1 wasexplored by Gene Set Enrichment Analysis (GSEA) software. The expression of MAST1 mRNA was low in glioma tissues. MAST1 mRNA expression was correlated with patient’s age, WHO grade, IDH mutation, and 1p/19q co-deletion status. Low expression of MAST1 was associated with poor prognosis in glioma patients. Further enrichment analysis revealed the involvement of MAST1 in multiple biological processes. It is evident that MAST1 is a potential indicator for prognostic evaluation in glioma patients, and this study provides a theoretical basis for understanding the pathogenesis of glioma.
