  • 摘要: 为了探究RNA甲基转移酶NSun2在胃癌发生发展中的生物学功能和作用机制,尝试利用构建的过表达和敲低NSun2的胃癌细胞系,运用蛋白质双向电泳联合质谱的方法,寻找其潜在的下游底物.实验结果显示,在体外NSun2可以促进胃癌细胞的增殖,蛋白质组学方法筛选并鉴定出34个差异蛋白,其中蛋白谷胱甘肽S转移酶P(GSTP1-1)和蛋白激酶C抑制蛋白1(KCIP-1)可能是NSun2在胃癌中的潜在作用靶点.这对于认识胃癌发生发展的分子机制具有重要意义.

     

    Abstract: To explore the role and function of RNA methyltransferase NSun2 in gastric cancer, NSun2 overexpression and knockdown gastric cancer cell lines were constructed, and a combination of protein two-dimensional electrophoresis and mass spectrometry analysis was done to identify potential NSun2 targets. NSun2 was found to promote the proliferation of gastric cancer cells in vitro. A total of 34 proteins were identified after proteomics analysis, including glutathione S-transferase P (GSTP1-1) and kinase C inhibitor protein 1 (KCIP-1), potential downstream targets of NSun2 in gastric cancer. Important insights into the molecular mechanisms underlying gastric cancer tumorigenesis and progression were gained.
